Pharmacology
Pharmacological action - anti-inflammatory, antipyretic, analgesic, gastroprotective.
Diclofenac - inhibits the synthesis of prostaglandins and has an anti-inflammatory, analgesic, antipyretic effect.
Misoprostol - suppresses basal and stimulated secretion of hydrochloric acid in the stomach, promotes the formation of bicarbonate, improves blood supply to the gastric mucosa.
Indications Diclofenac + Misoprostol
Osteoarthrosis, rheumatoid arthritis, ankylosing spondylitis.
Contraindications
Hypersensitivity, bleeding from the digestive tract (gastrointestinal tract).
Restrictions for use
Chronic heart failure, predisposition to fluid retention in the body, hepatic and / or renal failure.
pregnancy and lactation
Contraindicated in pregnancy and during breastfeeding.
When planning pregnancy, it should be borne in mind that misoprostol raises the tone of the uterus and can provoke a threat of miscarriage.
The use of diclofenac and other NSAIDs during pregnancy can cause premature closure of the arterial duct in the fetus, and as a consequence, hypertension in the small circulation.
The action category for the fetus by FDA is X.
Side effects of Diclofenac + Misoprostol
On the part of the intestine: nausea, diarrhea, gastralgia, dyspepsia, flatulence.
From the cardiovascular system and blood (hematopoiesis, hemostasis): reduced platelet aggregation, prolonged bleeding time, anemia (including hemolytic and aplastic), leukopenia, thrombocytopenia, eosinophilia, agranulocytosis.
From the nervous system and sensory organs: headache, dizziness.
Other: menorrhagia, metrorrhagia in women in the pre- and post-menopausal period, fluid retention, edema, skin rash.
Interaction
With simultaneous use with potassium-sparing diuretics, an increase in the K + content in the plasma (due to diclofenac) is possible. Diclofenac promotes the accumulation in the plasma of methotrexate, digoxin, lithium. Acetylsalicylic acid displaces diclofenac from binding sites, accelerates elimination and reduces the concentration of diclofenac. Myelotoxic drugs (drug) increase the manifestation of hematotoxicity of the drug.
