Mode of action - anti-allergic, antihistamine.

It blocks histamine H1 receptors.

Pharmacodynamics

Pharmacokinetics

Well absorbed from the digestive tract. The action begins in 30 minutes and lasts for 24 hours. It is metabolized in the liver with the participation of cytochrome P450 with the formation of an active metabolite - dexarboethoxyloratadine. Cmax of loratadine and its active metabolite in plasma are achieved after 1.3 and 2.5 hours, respectively. T1 / 2 loratadine - 8 h, dexarboethoxyloratadine - about 28 h.

Indications of the Loratadine 10-SL

Allergic rhinitis (seasonal, all-the-year-round), allergic rhinoconjunctivitis, chronic idiopathic urticaria, allergic itching dermatoses - contact allergic dermatitis, chronic eczema (as part of complex treatment).

Contraindications

Hypersensitivity (including its components), pregnancy, breast-feeding, children under 2 years.

Application in pregnancy and breastfeeding

Contraindicated in pregnancy. For the duration of treatment, breastfeeding should be stopped.

Side effects

From the nervous system and sensory organs: headache, fatigue, increased excitability.

On the part of the intestine: dryness of the oral mucosa, nausea, vomiting, gastritis.

Allergic reactions: skin rash.

Interaction

Erythromycin, cimetidine, ketoconazole increase the concentration in the plasma.

Dosing and Administration

Inside. Adults and children over 12 years of age - 10 mg once a day, daily dose - 10 mg. Children 2-12 years: body weight less than 30 kg - 5 mg once a day, daily dose - 5 mg, more than 30 kg - 10 mg once a day, daily dose - 10 mg. Overdose

Symptoms: drowsiness, tachycardia, headache.

Treatment: gastric lavage, administration of emetic and laxatives, activated charcoal.
