Cerebrolysin General Information

Cerebrolysin is a biotechnologically prepared peptide that stimulates neurotrophic regulation in the central nervous system.

Cerebrolysin is used worldwide for treatment of ischemic and hemorrhagic stroke, traumatic brain injuries, to treat different forms of dementia (vascular dementia, Alzheimer’s disease) and cognitive disorders and to prevent cognitive decline after brain injuries.

Cerebrolysin - Mode of Action

Cerebrolysin is a neurotrophic peptidergic drug with multimodal pharmacological properties indicated for the treatment of acute and chronic CNS disorders. The pathophysiological mechanisms targeted by Cerebrolysin belong to two distinct categories:

Pathological events and cascades caused by an injury or a degenerative disease and

Endogenous repair and recovery processes which constitute defense against an injury or progressing disease.

The natural repair and recovery processes are switched on within the CNS immediately after an injury. These processes also play important role in the continuous defense against progressing neurodegeneration characteristic of chronic CNS disorders (e.g. Alzheimer’s disease). Two major mechanisms regulate recovery from injuries at the molecular level: neurotrophic (NTF) and sonic hedgehog (Shh) signaling pathways. These signaling pathways regulate cellular processes of neurogenesis, angiogenesis, dendrite arborization, axonal sprouting, myelination, and remodeling of the neurovascular unit.

The pathological events and cascades after stroke or trauma lead to secondary injuries (additional loss of neural tissue and its interconnectivity) which further compromise motor and cognitive functions of a patient. Among the most relevant molecular processes targeted by Cerebrolysin in the acute phase of an injury are elements of the ischemic cascade, like excitotoxicity, uncontrolled apoptosis, overactivation of proteolytic enzymes, and overproduction of reactive oxygen species (ROS). In the early post-acute phase, lasting up to several weeks, Cerebrolysin prevents formation of toxic protein aggregates (amyloidosis), and lowers the level of inflammatory processes which further contribute to neurodegeneration.
